Product Introduction
The phosphamide technique is well suited to modify synthesized oligonucleotides with covalently linked reporter groups, linkers, spacers or haptens. 6-FAM, derived from the single isomer 6-carboxyl fluorescein, 5' -hexachlorofluorescein CE phosphamide (HEX) and 5' -tetrachlorofluorescein CE phosphamide (TET) can all be used to effectively label oligonucleotides.
Labeling of the 3' end of an oligonucleotide with fluorescein can be achieved using 3' -fluorescein CPG. When any of these carriers are used, standard cleavage and deprotection of fluorescein labeled oligonucleotides are performed with ammonium hydroxide. Since fluorescein phosphoamides do not contain dmt groups, demethylation steps are not required. After ammonia evaporation, the labeled oligonucleotides can be used for most applications.
