Oligonucleotide was coupled to Quasar 705 phospholamide by solid-phase chemical column. After capping and oxidation, the oligonucleotide is cleaved and deprotected to obtain a 5' labeled Quasar 705 final product.
Quasar 705 phospholamide was used for 5' -end labeling of oligonucleotide synthesis, which is suitable for a variety of applications, including double-labeled fluorescent probes for real-time PCR. Quasar 705 is an indocyanine dye that fluoresses in the dark red region of the visible spectrum and can be quenched effectively with either BHQ-2 or BHQ-3 dyes.
