　　1) coupling

　　UltraMild monomer and Cap A were routinely used. Oxidation with 0.5M CSO acetonitrile solution required 3min oxidation time.

　　2) Deprotection conditions

　　Oligonucleotides were lysed and deprotected with 30% ammonium hydroxide for 2 h at room temperature. If oxidized with ordinary monomer and iodine, better results can be obtained by deprotection with NH4OH/EtOH 3:1 (v/v) at 55°C for 17 hours.

　　The product description

　　MGB Eclipse is a combination of a small groove conjugate (MGB) tripeptide and a dark quencher. MGB significantly enhances the hybridization reaction, while Eclipse has a broad absorption range that quenches many of the most common fluorophores, including FAM, HEX, TET, and Yakima Yellow. MGB Eclipse is an important group for the synthesis of hydrolytic probes, which are the most common probe detection method for qPCR.
