Instructions for use:
1) coupling
The recommended coupling time is 3 minutes
2) Take off protection
Oligonucleotides containing AquaPhluor 593 can be deprotected with ammonium hydroxide, and excellent results can be obtained under the following conditions:
- 2 hours at 65°C (optimal)
- 17 hours at room temperature (optimal)
- 17 hours at 55°C (preferred)
Note: When using fast lytic carriers, deprotection with ammonium hydroxide may result in side reactions that produce small amounts of non-fluorescent impurities. To avoid the appearance of impurities, the carrier can be treated with a blocked base (e.g., 10% diethylamine ACN solution) for 2-3 minutes prior to lysis/deprotection.
